Abstract:

A flavour enhancer that is frequently utilized is monosodium glutamate (MSG). This may result in kidney injury due to oxidative stress. Nigella sativa oil (NSO) and zinc oxide nanoparticles (ZnO-NPs) have the potential to exert renoprotective effects. Aim: This study was performed to compare between protective effects ZnO-NPs or NSO alone and combined on renal toxicity induced by MSG. Methods: There were five groups of 60 healthy adult albino male rats. Group 1 was the control group, and Group 2 received MSG orally at a dosage of 200 mg/kg bw/day. Group 3 was administered MSG orally and ZnO-NPs intraperitoneally at a rate of 5 mg/kg bw/day. Group 4 was administered MSG with NSO orally at a rate of 2 ml/kg bw/day. Group 5 received MSG along with ZnO-NPs and NSO. All medications were administered for a period of four weeks. Kidney samples were processed for light and electron microscopic examinations at the end of the experiment. Results: MSG-treated rats had histological and ultrastructural kidney damage including shrunken glomeruli, widened Bowman’s space, tubular degeneration and vacuolated cytoplasm. The ZnO-NPs group showed moderate protection with nearly normal glomeruli and minimal tubular changes. The NSO group showed mild protection with partial preservation of glomerular and tubular structures. Combined treatment with ZnO-NPs and NSO resulted in the best results with kidneys appearing nearly normal and closely similar to control group. Conclusion: Combined use of ZnO-NPs and NSO improved kidney structure in MSG-induced toxicity rats.
